Synthesis of 2′‐Deoxy‐2′‐fluoro‐ <scp>L</scp> ‐cytidine and Fluorinated <scp>L</scp> ‐Nucleic Acids for Structural Studies

The unique properties of fluorine atom on nucleic acid backbone can offer stricking functional and structural features. In order to extend the biological applications L-type acid, we chemically incorporate fluoro-modification into 2′-position L-cytidine, obtain a series fluoro-modified L-DNAs/ L-RNAs. Our melting study indicates that 2′-fluoro-modification does not disrupt thermostabilities wild-type L-nucleic acids. Consistently, our X-ray crystal structure reveals fluoro-moiety cause no perturbation, fluoro-L-cytidine forms Watson-Crick base pair with L-guanosine virtually identical nonmodified CG pair. This cytidine provides useful biochemical strategy investigate as advanced molecular therapy.
